as the antigen for a simplified precipitation test for syphilis. Cardiolipin is the lipid which, in combination with lecithin, gives the alcoholic extract of beef heart its antigenic properties. It is a complex phosphatidic acid with a molecular weight of 2195 and iodine number 120-125. It requires lecithin and cholesterol for antigenicity. It has not yet been synthesized. The cardiolipin-lecithin antigen may be adjusted to any desired sensitivity, within limits, merely by altering the ratio of cardiolipin and lecithin. Kline (1947) recommended a cardiolipin-lecithin ratio of 1: 9 4 and 1: 10 6. Our attempts to isolate and prepare the pure cardiolipin from beef heart were not successful. We have used as our antigen a cardiolipin-lecithin mixture of 1 : 10, standardized by Dr. Kline and supplied by the La Motte Chemical Company. The cardiolipin-lecithin (C.L.) test is now done as

The Harris Slide Test: A Microflocculation Test for Syphilis with Cardiolipin Antigen.

The antigens in common use for the serological diagnosis of syphilis, such as the complement fixation tests and the various flocculation tests, are alcoholic extracts of ox heart usually sensitized with cholesterol. If, however, cholesterol is added to the crude alcoholic extract, false positive reactions are not uncommon. On this account it has become customary to purify the alcoholic extracts...

VDRL slide flocculation reaction for syphilis performed on active sera.

The discovery of cardiolipin (Pangborn, 1941a) as the active agent in antigens for sero-reactions for syphilis enabled the production of chemically defined antigens, composed of cardiolipin, purified lecithin (Pangborn, 1941b), and cholesterol. Among the many tube and slide flocculation reactions based on the use of cardiolipin antigens, the Venereal Disease Research Laboratory (VDRL) slide flo...

The VDRL tube flocculation test; a microflocculation test for syphilis using cardiolipin-lecithin-cholesterol antigen.

In an earlier paper in this Journal (Vogelsang, 1947), a new microflocculation test for syphilis described by Harris, Rosenberg, and Riedel (1946) was compared with the Wassermann reaction, the Kahn standard test, and the Meinicke clarification test in 5,556 sera. The test was found to be very useful in the diagnosis of syphilis, having a satisfactory sensitivity and a good specificity. Harris ...

Serological, nephelometrical, and statistical studies on the employment of synthetic lecithin in cardiolipin antigens.

Cardiolipin antigens, used in the sero-diagnosis of syphilis, are mixtures of the phospholipids, cardiolipin, and purified natural lecithin (beefheart or egg), and usually also contain cholesterol. In 1949 Baer and Kates (1949, 1950) succeeded in the synthesis of L-ac-dimyristoyl lecithin and in the following years other types of synthetic lecithin were produced, including a D-oc-dimyristoyl le...
